Significance is in the eye of the reader. I would say it is significant.
Maybe you could describe it as someone saying what he knows to be true, but wants it confirmed explicitly just to be sure.

On September 21st, Frodo and Sam set out together. Sam thought he wast setting Frodo towards Rivendell to see Bilbo. But, of course, Bilbo (with Elrond and Galadriel) met them the next day and that is when Sam realized that Frodo was not going to Rivendell, but to the Havens.
